To investigate the expression of large tumor suppressor homolog 2 (LATS2) gene and its promotor methylation in oral squamous cell carcinoma (OSCC). Reverse transcription-PCR (RT-PCR) and pyrosequencing were used to detect the mRNA and promotor methylation of LATS2 gene in 72 OSCC specimens and normal oral mucosa tissues. Western blotting was used to detect the LATS2 protein in six OSCC specimens and normal oral mucosa tissues. All cases had expression of LATS2 mRNA in normal oral mucosa tissues, but the expression was down-regulated significantly, only 47% (34/72) in 72 cases of OSCC showed LATS2 mRNA expression. The expression was correlated with the degree of tumor differentiation and lymph node metastasis (0.05). The results of pyrosequencing show that 68% of promotor methylation (49/72) in 72 cases of OSCC. Furthermore, there was significant correlation between the mRNA and promotor methylation of LATS2 gene (χ(2)=16.980, 0.01). All the six specimens had the low LATS2 protein expression. The promotor methylation of LATS2 gene may play an important role in the occurrence of OSCC.
探讨大肿瘤抑制因子同源物2(LATS2)基因在口腔鳞状细胞癌(OSCC)中的表达及其启动子甲基化情况。采用逆转录聚合酶链反应(RT-PCR)和焦磷酸测序法检测72例OSCC标本及正常口腔黏膜组织中LATS2基因的mRNA及启动子甲基化情况。采用蛋白质免疫印迹法检测6例OSCC标本及正常口腔黏膜组织中LATS2蛋白的表达情况。所有病例的正常口腔黏膜组织均有LATS2 mRNA表达,但表达明显下调,72例OSCC中仅47%(34/72)有LATS2 mRNA表达。其表达与肿瘤分化程度及淋巴结转移相关(P<0.05)。焦磷酸测序结果显示,72例OSCC中有68%(49/72)存在启动子甲基化。此外,LATS2基因的mRNA与启动子甲基化之间存在显著相关性(χ²=16.980,P<0.01)。6例标本的LATS2蛋白表达均较低。LATS2基因的启动子甲基化可能在OSCC的发生中起重要作用。
