Abstract

BACKGROUND

Currently, mushrooms have been used in traditional and folk medicines for their therapeutic activities, such as antibiotic, antitumor, anti-inflammatory, anticancer, antileukemic and immunomodulatory actions. This investigation evaluates the anti-invasive, antiproliferative and cytotoxic effects of Pleurotus ostreatus (Pleurotaceae) on leukemia cell lines.

METHODS

The proliferation of KG-1 cells was measured by using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay after treatment with gradient dilutions of P. ostreatus extract. Then, the minimum inhibitory concentration (MIC) of the extract was determined. Moreover, the proliferation of Jurkat cells and bone marrow mesenchymal stem cells (BMSCs), a cancerous cell line and normal body cells, respectively, was considered. The apoptotic morphology of treated KG-1 cells was evaluated with Giemsa staining. The invasion and migration of cells were evaluated using transwell invasion assay. Thereafter, the rates of apoptosis and necrosis were measured by using flow cytometry, and BAX and MMP-9 gene expression were evaluated using quantitative reverse transcription-polymerase chain reaction as apoptotic and metastatic genes, respectively.

RESULTS

The MIC of the extract was determined to be 1 mg/mL after 48 h. According to the results, the extract decreased the proliferation of leukemia cell lines (KG-1 and Jurkat cells) but had no antiproliferative effects on BMSCs. Moreover, KG-1 cell migration and MMP-9 gene expression decreased after the treatment, and the rate of apoptosis and BAX gene expression increased significantly.

CONCLUSIONS

According to the efficient therapeutic properties of P. ostreatus on leukemia cell lines, this mushroom could be introduced as a natural medicine to cure leukemic patients who suffer from the harmful side effects and enormous costs of chemotherapy.

目前,蘑菇因其治疗活性已被用于传统医学和民间药物中,如具有抗生素、抗肿瘤、抗炎、抗癌、抗白血病和免疫调节作用。本研究评估了平菇(侧耳科)对白血病细胞系的抗侵袭、抗增殖和细胞毒性作用。

方法

用平菇提取物梯度稀释液处理后,采用3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐法检测KG-1细胞的增殖情况。然后,测定提取物的最低抑菌浓度(MIC)。此外,还分别考察了白血病癌细胞系Jurkat细胞和正常体细胞骨髓间充质干细胞(BMSC)的增殖情况。用吉姆萨染色评估处理后KG-1细胞的凋亡形态。采用Transwell侵袭实验评估细胞的侵袭和迁移能力。此后,用流式细胞术检测凋亡和坏死率,并用定量逆转录-聚合酶链反应分别评估凋亡基因BAX和转移基因MMP-9的表达。

结果

48小时后提取物的MIC测定为1mg/mL。结果显示,提取物可降低白血病细胞系(KG-1和Jurkat细胞)的增殖,但对BMSC无抗增殖作用。此外,处理后KG-1细胞迁移和MMP-9基因表达降低,凋亡率和BAX基因表达显著增加。

结论

鉴于平菇对白血病细胞系具有有效的治疗特性,这种蘑菇可作为一种天然药物用于治疗白血病患者,这些患者因化疗产生有害副作用且费用高昂。
