This paper aims to explore the expression characteristics of mitochondrial function-related genes in patients with first-episode schizophrenia (SCZ)and the correlation between differentially expressed genes and clinical metabolic indicators.
Twenty patients with first-episode SCZ who had not taken antipsychotic drugs (patient group) and twenty healthy controls (control group) were included. Quantitative real-time PCR technology was used to detect the expression levels of genes related to mitochondrial quality control and oxidative phosphorylation in peripheral blood leukocytes, and metabolic indicators such as blood biochemistry and blood glucose were collected.
The gene expression levels of key genes related to mitochondrial function, PGC-1a, PARK2, and LC3B, in the patient group were significantly lower than those in the control group (P < 0.05). Correlation analysis showed that the expression level of PGC-1a gene in the patient group was negatively correlated with very low-density lipoprotein levels (r =-0.451), and the expression level of PARK2 gene in the patient group was negatively correlated with uric acid levels (r =-0.447).
The expression levels of multiple key genes in the mitochondrial quality control and oxidative phosphorylation processes in patients with first-episode SCZ display a downward trend. The differentially expressed genes are correlated with the metabolic abnormalities of the patients, suggesting that mitochondrial dysfunction may be related to the high incidence of metabolic diseases in patients with SCZ.
探讨首发精神分裂症(SCZ)患者线粒体功能相关基因的表达特征,以及差异表达基因与临床代谢指标之间的相关性。
纳入20例未服用抗精神病药物的首发SCZ患者(患者组)和20例健康对照者(对照组)。采用定量实时PCR技术检测外周血白细胞中线粒体质量控制和氧化磷酸化相关基因的表达水平,并收集血液生化、血糖等代谢指标。
患者组中线粒体功能关键基因PGC-1a、PARK2和LC3B的基因表达水平显著低于对照组(P<0.05)。相关性分析显示,患者组中PGC-1a基因的表达水平与极低密度脂蛋白水平呈负相关(r=-0.451),患者组中PARK2基因的表达水平与尿酸水平呈负相关(r=-0.447)。
首发SCZ患者线粒体质量控制和氧化磷酸化过程中多个关键基因的表达水平呈下降趋势。差异表达基因与患者的代谢异常相关,提示线粒体功能障碍可能与SCZ患者代谢疾病的高发有关。
