Tumor is a complex disease that seriously threatens human health. Traditional treatment methods have many limitations. In recent years, with the in-depth study of tumor immune mechanisms, the mRNA vaccine, as an innovative immunotherapy strategy, has attracted wide attention due to its unique advantages and great potential. The purpose of this review is to introduce the technical development status of tumor mRNA vaccines, the application progress in a variety of tumors, and the challenges and future development directions, to provide reference for related research. In this paper, we reviewed the recent literature on tumor mRNA vaccines, analyzed the technical progress in structure modification, delivery tools, immunogenicity regulation, and summarized its application in different tumor types, and discussed the current challenges and future development directions. Conclusions: Remarkable progress has been made in structural modification, delivery tool optimization, and immunogenicity regulation of tumor mRNA vaccines, which show good application prospects in the treatment of a variety of tumors. However, the complexity of the tumor microenvironment, the limitation of immune activation, the stability and translation efficiency of mRNA, and the optimization of delivery systems are still challenges. In the future, the application of new technologies, the exploration of combined treatment strategies, and the advancement of personalized medicine will provide new opportunities for the development of tumor mRNA vaccines. With the continuous progress of technology, tumor mRNA vaccines are expected to play an important role in tumor treatment and bring greater well-being to human health.
肿瘤是一种严重威胁人类健康的复杂疾病。传统治疗方法存在诸多局限性。近年来,随着对肿瘤免疫机制研究的深入,mRNA疫苗作为一种创新的免疫治疗策略,因其独特优势和巨大潜力而备受关注。本综述旨在介绍肿瘤mRNA疫苗的技术发展现状、在多种肿瘤中的应用进展以及面临的挑战和未来发展方向,为相关研究提供参考。本文回顾了近期关于肿瘤mRNA疫苗的文献,分析了其在结构修饰、递送工具、免疫原性调控方面的技术进展,总结了其在不同肿瘤类型中的应用,并探讨了当前面临的挑战和未来发展方向。结论:肿瘤mRNA疫苗在结构修饰、递送工具优化和免疫原性调控方面取得了显著进展,在多种肿瘤治疗中显示出良好的应用前景。然而,肿瘤微环境的复杂性、免疫激活的局限性、mRNA的稳定性和翻译效率以及递送系统的优化仍是挑战。未来,新技术的应用、联合治疗策略的探索以及个性化医疗的推进将为肿瘤mRNA疫苗的发展提供新机遇。随着技术的不断进步,肿瘤mRNA疫苗有望在肿瘤治疗中发挥重要作用,为人类健康带来更大福祉。
