To explore the improvement effect of electroacupuncture (EA) based on acupuncture (acupuncture for regaining consciousness and opening orifices) on cognitive impairment in mice with Parkinson's disease (PD), and to explore its regulatory mechanisms on the kinesin family member 5A (KIF5A)/mitochondrial Rho GTPase 1 (Miro1) pathway and mitophagy in prefrontal cortical neurons.
A total of 70 male C57BL/6J mice of clean grade were randomly divided into a normal group (12 mice), a sham operation group (12 mice), and a model pre-screening group (46 mice). Unilateral stereotaxic injection of 6-hydroxydopamine (6-OHDA) into the medial forebrain bundle was adopted to establish the PD model in the model pre-screening group. Twenty-four mice after successful modeling were randomly selected and divided into a model group and an EA group, 12 mice in each one. In the EA group, acupuncture was applied at "Shuigou" (GV26) and bilateral "Sanyinjiao" (SP6) and "Neiguan" (PC6), ipsilateral "Sanyinjiao" (SP6) and "Neiguan" (PC6) were connected to EA respectively, with disperse-dense wave, 5 Hz/20 Hz in frequency, 0.5 mA in current intensity, 20 min a time, 6 times a week for 30 days. Cognitive function was assessed by Y-maze and Morris water maze tests; morphology of prefrontal cortex was observed by H.E. staining; reactive oxygen species (ROS) level in prefrontal cortex was detected by fluorescence probe method; mitochondrial morphology and autophagosome ultrastructure were observed by transmission electron microscopy; the mRNA expression of tyrosine hydroxylase (TH) was detected by quantitative real-time PCR; the protein expression of TH, KIF5A, Miro1, p62, Parkin and PTEN induced kinase 1 (PINK1) was detected by Western blot.
Compared with the sham operation group, both the model group and the EA group exhibited increased rotation number of per minute (<0.001). Compared with the sham operation group, in the model group, the novel arm exploration time of Y-maze test was shortened (<0.001), the escape latency of Morris water maze test was prolonged (<0.05) and the platform crossing number of Morris water maze test was reduced (<0.01); in the prefrontal cortex, the number of cellular vacuole and neurons with karyopyknosis was increased (<0.001), and mitochondrial autophagosomes could be observed; in the prefrontal cortex, the relative expression of ROS was increased (<0.001), the protein and mRNA expression of TH was decreased (<0.001), the protein expression of Miro1, PINK1, Parkin was increased (<0.001, <0.01), the protein expression of KIF5A and p62 was decreased (<0.001). electroacupuncture effectively alleviates cognitive impairment and damage of neuronal function in PD mice, its mechanism may be related to the regulation of KIF5A/Miro1 pathway, hence reducing the mitophagy in prefrontal cortical neurons.
探讨基于醒脑开窍针法的电针(EA)对帕金森病(PD)小鼠认知功能障碍的改善作用,并探讨其对前额叶皮质神经元中驱动蛋白家族成员5A(KIF5A)/线粒体Rho GTP酶1(Miro1)通路及线粒体自噬的调控机制。
将70只清洁级雄性C57BL/6J小鼠随机分为正常组(12只)、假手术组(12只)和模型预筛组(46只)。模型预筛组采用单侧立体定向注射6-羟基多巴胺(6-OHDA)至内侧前脑束建立PD模型。将造模成功后的24只小鼠随机分为模型组和EA组,每组12只。EA组针刺“水沟”(GV26)及双侧“三阴交”(SP6)、“内关”(PC6),患侧“三阴交”(SP6)和 “内关”(PC6)分别连接电针,采用疏密波,频率为5Hz/20Hz,电流强度0.5mA,每次20min,每周6次,共30天。采用Y迷宫和Morris水迷宫试验评估认知功能;采用苏木精-伊红(H.E.)染色观察前额叶皮质形态;采用荧光探针法检测前额叶皮质活性氧(ROS)水平;采用透射电子显微镜观察线粒体形态和自噬体超微结构;采用实时定量聚合酶链反应检测酪氨酸羟化酶(TH)的mRNA表达;采用蛋白质免疫印迹法检测TH、KIF5A、Miro1、p62、Parkin和PTEN诱导激酶1(PINK1)的蛋白表达。
与假手术组比较,模型组和EA组每分钟旋转次数均增加(<0.001)。与假手术组比较,模型组Y迷宫试验新臂探索时间缩短(<0.001),Morris水迷宫试验逃避潜伏期延长(<0.05),Morris水迷宫试验平台穿越次数减少(<0.01);前额叶皮质细胞空泡及核固缩神经元数量增加(<0.001),可见线粒体自噬体;前额叶皮质ROS相对表达增加(<0.001),TH蛋白和mRNA表达降低(<0.001),Miro1、PINK1、Parkin蛋白表达增加(<0.001,<0.01),KIF5A和p62蛋白表达降低(<0.001)。与模型组比较,EA组Y迷宫试验新臂探索时间延长(<0.01),Morris水迷宫试验逃避潜伏期缩短(<0.05),Morris水迷宫试验平台穿越次数增加(<0.05);前额叶皮质细胞空泡及核固缩神经元数量减少(<0.001),线粒体自噬体数量减少,线粒体形态改善;前额叶皮质ROS相对表达降低(<0.01),TH蛋白和mRNA表达增加(<0.001,<0.01),Miro1、PINK1、Parkin蛋白表达降低(<0.001,<0.01,<0.05),KIF5A和p62蛋白表达增加(<0.01,<0.05)。
电针可有效减轻PD小鼠的认知功能障碍及神经元功能损伤,其机制可能与调控KIF5A/Miro1通路,从而减少前额叶皮质神经元的线粒体自噬有关。
