Abstract

Circular RNA (circRNA), a type of noncoding RNA, features a covalently closed circular structure lacking 5'-3' polarity and a polyadenylated tail. Its resistance to RNase R degradation endows it with greater stability and resilience than linear RNA, leading to a prolonged half-life. Recent research has highlighted the crucial role of circRNAs, especially circPRMT5, in diverse cancers. CircPRMT5 is generated by reverse splicing of exon 18 within a 682-nucleotide gene. In various cancer cells, elevated circPRMT5 expression is correlated with increased tumor cell proliferation, migration, and invasion abilities. It impacts tumor progression by regulating cell proliferation, migration, invasion, and chemotherapy resistance. This review comprehensively examines the expression patterns, mechanisms, and clinical potential of circPRMT5 in cancers such as breast, bladder, nonsmall cell lung, esophageal, thyroid, liver, colorectal, and gastric cancer. Overall, circPRMT5 shows promise as a biomarker and therapeutic target, providing valuable insights for future cancer research and treatment strategies.
